How it works
PureLink™ Pro Quick96 Plasmid The kit uses two 96-well plates, a filter plate (clear O-ring), and a binding plate (red O-ring) to purify plasmid DNA. Bacterial cells were alkaline lysed and pelleted by centrifugation. Bacterial lysates are then spread onto Quick96 filter plates for clearance and collection in Quick96 binding plates, where plasmid DNA is bound to the silica membrane. After removing contaminants through washing steps, the purified DNA is eluted and ready for downstream applications. The entire procedure can be completed in 45 minutes (Figure 1).

Improved Plate Design
PureLink™ Pro Quick96 Plasmid Kit offers an optimized semi-skirted 96-well plate for improved performance and results (Figure 2). Quick96 filter plates are designed for rapid and efficient removal of bacterial lysate. Quick96 binding plates provide enhanced binding capacity for binding up to 20 µg of plasmid DNA. In addition, the plate nozzle containing an annular collar prevents sample cross-contamination and improves drying of the silica membrane, thereby preventing ethanol residue. Alternatively, samples can be processed manually via centrifugation or via an automated liquid handling platform using a benchtop vacuum manifold. The semi-skirted panel design is compatible with most vacuum manifolds on automated workstations.

Higher Yields and Purity
PureLink™ Pro Quick96 Plasmid Kit provides fast and reproducible purification of plasmids from 5 mL LB, 3 mL 2xYT or 3 mL 20 µg of plasmid DNA from an E. coli strain grown in TB. In the automated platform, using 1.5 mL of E. coli culture grown overnight in square-well modules, typical yields were 5-15 µg of plasmid DNA, with an average of 9.4 µg (Figure 3). The resulting plasmid DNA was supercoiled, and no genomic DNA or RNA was detected (Figure 4). PureLink™ Pro Quick96 Plasmid Kit provides higher yields of high-quality plasmid DNA for common downstream applications such as automated sequencing, PCR, restriction enzyme digestion, and cloning.
